Is it so easy to do a DNA test. Just pluck hair from 2 random person and just do the test. Can someone pls explain me how to do DNA as I have no idea how is it done. As I have seen in movies I usually see that court order for such test and the report is very confidential. I also have watched in one movies doctor getting signature before doing test . So someone from medical field pls explain me how is it done and where is it done? If the repor is confidential are the supposed to write the name of person undergoing on the envelope ? Am just curious to know about the procedure of the test as now a days we hear about such test more often...

i too have the same doubt yaar..
i thought that some legal procedures are involved for doing a dna test..
i didnt know that anyone can do this if we have two hairs from 2 people..
is this true or imagination of cv's??god knows yaar..anything can happen in ipk

I think it's not that easy...There is some legal procedure and it takes some time...and expensive...

paternity test was being done ... in simple words yeah hair plucked frm roots can to be used to obtain DNA ... but for 100% accurate one hair is not enough .. swab of mouth is suited or blood .. it takes weeks to obtain result ... my knowledge comes frm various forensic shows on US tv .. any doctor on the forum can give better know how

Well, I think things are not so difficult in India! I don't think there are as many legal hassles!

well,
hair has a protein calld karotene...The structure of the hair is composed of three layers as follows:
  • The cortex
  • Cuticle
  • Medulla
DNA in hair is available only in the root of the hair!
DNA Extraction from hair:
Cut 10- 15 hair roots about 0.5 cm into a 1.5ml eppendorf tube.
Add 50 ul of 200mM NaOH solution.
Boil the tube in a water bath at 94oC for 10 minutes.
Cool at room temperature and add 50 ul of a solution containing 200mM HCL.
+100mM Tris-HCL pH 8.5.

after this procedure...dna is perfectly extracted...the samples are ready for something called the PCR (polymerase chain reactin) or RFLP...(restriction fragment length polymorphism)

PCR is to duplicate sequences if there is very less sample!

the percent of similarities will determine the relation ships between people!!

this is how an rflp map looks!!
